METHODS: The composites of ICPC/FS/bBMP, ICPC/FS or bBMP were injected subcutanously or implanted into thigh muscle pouches of 45 mice at random.The effect of ectopic bone formation was evaluated by biochemical examination of alkaline phosphatase (ALP) and histological examination 2 and 4 weeks after operation and the new bones were weighed in the 4th postoperative week to compare statistically the ability of bone formation of different composites. RESULTS: ICPC/FS/bBMP composite induced the formation of new bone most effectively. BBMP stimulated cess osteogenesis while ICPC/FS showed no sign of new bone formation.There was a significant difference among them (P<0.05). CONCLUSION: The injectable bone substitute, ICPC/FS/BMP, exhibited an excellent osteoinductive activity,and ICPC/FS was a good carrier for BMP. 【Keywords】 injectio; calcium phosphates; bone cements; fibrin tissue adhesive; bone morphogenetic proteins; carrier; osteoinduction 【摘要】目的: 了解以注射型磷酸钙骨水泥(ICPC)/纤维蛋白胶(FS)共同作为骨形态发生蛋白(BMP)复合载体的注射型骨修复材料在小鼠体内异位成骨的能力,探讨其作为注射性骨替代物的可行性. 方法: 在小鼠肌袋随机注射或植入ICPC/FS/bBMP复合物、ICPC/FS复合物和单纯bBMP材料,术后2 wk, 4 wk取材,通过碱性磷酸酶测定和组织学检查,观察其异位诱导成骨能力. 并对术后4 wk时新生骨进行定量测定和统计学比较. 结果: ALP和组织学检查结果表明ICPC/FS/bBMP复合物有较多新骨形成,单纯bBMP诱导出少量骨,ICPC/FS复合物未见新生骨,诱导骨量有统计学意义(P<0.05). 结论: 可注射性ICPC/FS/BMP复合物具有良好的骨诱导活性,ICPC/FS是BMP的良好注射性载体之一. 【关键词】 注射剂;磷酸钙类;骨粘合剂;纤维蛋白组织粘着剂;骨形态发生蛋白质类;载体;骨诱导 开放植骨是植骨术的传统方式,但近年来随着微创伤外科技术的发展,研制和应用可注射性骨替代材料已成为一个重要的努力方向[1-2]. 注射型磷酸钙骨水泥(injectable calcium phosphate cement, ICPC)具有良好的生物相容性和根据骨缺损形状临时塑性的特性,使其成为目前最具潜力的植骨材料之一. 但其空隙率低,降解缓慢,影响新骨的长入[3-4]. 而纤维蛋白胶(fibrin sealant, FS)不仅具有良好的止血、粘结功能和可注射性,而且生物相容性良好、易于被人体吸收, 是骨形态发生蛋白(bone morphogenic protein, BMP)的理想载体[5-6],已广泛用于许多外科领域. 我们以研制具有骨诱导、可塑形和能降解的注射型骨移植材料为目标,在ICPC中加入降解快的FS,组成注射型复合载体,可望提高ICPC的降解速度,加速新骨的生长. 1材料和方法 1.1材料 1.1.1以ICPC/
